As a serine/threonine protein kinase, ILK connects integrin-mediated cell-matrix interactions and downstream signaling pathways, including the PI3K/Akt and Wnt pathways. It is involved in a variety of physiological processes, such as embryonic development, tissue homeostasis, and angiogenesis, while its dysregulation is associated with pathological states such as cancer metastasis, fibrosis, and cardiovascular disease. The ILK gene encodes a multidomain protein that interacts with multiple proteins, including PINCH, parvins, and β-integrin, to form an IPP complex (ILK-PINCH-Parvin), thereby regulating cytoskeletal dynamics and focal adhesion assembly.
