| Summary | Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance. |
| Sterility | Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination. |
| Ad5 E1 Detection | All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination. |
| RCA Assays | Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources. |
| PFU Titering | All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells. |

The CLDN3 gene encodes claudin-3 (CLDN3), which belongs to the tight junction protein family and plays a key role in maintaining cell polarity, barrier function, and intercellular communication. Claudin is an integral membrane protein that forms the backbone of tight junctions and selectively regulates paracellular permeability and ion transport across the epithelium and endothelium. CLDN3 is particularly abundant in tissues such as the kidney, liver, and gastrointestinal tract, where it helps maintain tissue integrity and fluid homeostasis. Dysregulation of CLDN3 has been implicated in a variety of pathological conditions, including cancer, and its overexpression or inhibition can affect tumor progression, metastasis, and drug resistance. Due to its involvement in cell adhesion and signaling pathways, CLDN3 has emerged as a potential therapeutic target and biomarker for diseases characterized by epithelial barrier dysfunction.
Human CLDN3 adenoviral particles are engineered viral vectors designed to deliver the CLDN3 gene to target cells for research or therapeutic purposes. These particles are based on an adenovirus serotype (usually Ad5) that has been modified to be replication-defective, ensuring safety while maintaining high transduction efficiency in a variety of cell types, both dividing and non-dividing. These particles are widely used in functional studies to investigate the role of CLDN3 in tight junction formation, epithelial barrier regulation, and disease mechanisms. In addition, they can serve as a tool for gene therapy studies, especially in diseases where CLDN3 modulation may restore normal cellular function.
