Wild-type human adenovirus type 30 (HAdV-30) is a non-enveloped, double-stranded DNA virus belonging to the genus Mastadenovirus within the family Adenoviridae. Its transmission routes primarily include respiratory droplet transmission, fecal-oral transmission (especially in outbreaks associated with contaminated water sources or poor sanitation), and contact with contaminated surfaces. HAdV-30 exhibits tropism for respiratory and gastrointestinal epithelial cells, leading to localized infections. The virus enters cells via receptor-mediated endocytosis (typically utilizing the coxsackievirus and adenovirus receptor CAR or CD46), uncoats in the cytoplasm, and delivers its genome to the nucleus. Viral replication occurs in the nucleus, causing cytopathic effects, including cell rounding and detachment. Its pathogenic mechanisms include direct lysis of infected cells and induction of inflammatory responses, leading to the manifestation of symptoms.
The HAdV-30 virion has a typical icosahedral capsid structure, approximately 70-90 nanometers in diameter, composed of 252 capsomeres: 240 hexons (trimers of hexon protein) forming the triangular faces and edges, and 12 pentons (pentamers of penton base protein) located at the vertices. Each penton base is attached to a protruding, globular fiber protein trimer, which is crucial for the virus''s binding to host cell receptors. The genome consists of a linear double-stranded DNA molecule approximately 36 kilobase pairs (kbp) long, flanked by inverted terminal repeat sequences (ITRs) that serve as origins of replication. It encodes numerous early genes (E1A, E1B, E2, E3, E4) and late genes (L1-L5) as well as structural proteins. Early genes regulate viral DNA replication, modulate the host immune response (e.g., E3 proteins interfere with MHC-I presentation and TNF-mediated apoptosis), and alter the cellular environment. Late genes primarily encode structural capsid proteins (hexon protein, penton base protein, fiber protein) and proteins involved in virion assembly. The viral genome also contains viral-associated RNAs (VA RNAs), which can counteract the host''s interferon response. Serotype classification primarily depends on the antigenic variations in the hexon protein and the hypervariable region of the fiber protein head.
