Description Leveraging two major technology platforms—KASP (Kompetitive Allele-Specific PCR) and TaqMan MGB probes—we provide cost-effective or ultra-high-specificity custom genotyping services for any species and any genetic locus.
Introduction If the customer provides a specific genotyping RS number, we can perform custom development and offer SNP testing services upon receipt of mailed blood/tissue/cell samples. The customer needs to provide: 1. Sample materials such as cells (≥10^6 cells), tissue (≥300 mg), blood (≥1 ml), serum (≥1.5 ml), etc.; nucleic acid extraction is charged separately; 2. Genomic DNA (volume ≥30 μl, concentration ≥50 ng/μl), with purity OD260/280 between 1.7 and 1.9; 3. For the human genome, the RS number of the SNP locus must be provided. For other species without rs numbers, such as cattle, chickens, and fish, the exact 200 bp sequences flanking both upstream and downstream of the SNP locus must be provided, along with the mutation type at the SNP locus and whether any other linked loci exist within 25 bp upstream or downstream of the locus. We deliver: 1. SNP results (Excel spreadsheet); 2. Complete experimental report: amplification and reaction systems, primers involved, and probe sequences; Scatter plot of clustering results (KASP method) / Amplification curves and Ct value analysis (TaqMan MGB method). 3. Other relevant materials required by the customer.

Selenoprotein S (Selenoprotein S, SELENOS/SEPS1) is an important selenium-dependent protein mainly localized to the endoplasmic reticulum membrane, participating in key biological processes such as endoplasmic reticulum stress regulation, misfolded protein degradation, inflammatory response regulation, and antioxidant protection. The -105G/A locus upstream of the SELENOS gene promoter region is the most extensively studied important functional single nucleotide polymorphism (SNP) of this gene, with G/A and G/C alleles, where G is the wild-type allele and A and C are mutant alleles. Studies have found that this SNP locus regulates SELENOS gene transcriptional activity by affecting the integrity of transcription factor binding sites; G allele carriers have higher SELENOS expression levels, stronger ER stress regulatory capacity, and lower inflammatory responses, whereas A allele carriers show reduced SELENOS expression and increased pro-inflammatory cytokine production, forming a chronic inflammatory state. The AA/GA genotypes are significantly associated with susceptibility to various diseases, including infectious diseases (brucellosis, EV71 virus infection), malignant tumors (gastric cancer), cardiovascular diseases (coronary heart disease, ischemic stroke), and metabolic diseases (type 2 diabetes, gestational diabetes), with A allele carriers showing a 1.3- to 1.8-fold increased disease risk. Allele frequencies vary by ethnicity; among the Chinese Han population, the G allele frequency is approximately 0.60-0.65 and the A allele frequency approximately 0.35-0.40. In addition, this locus exhibits interaction effects with environmental factors such as selenium nutritional status and metal exposure, and AA genotype individuals may benefit more from selenium supplementation interventions. Research on this promoter locus provides an important molecular marker for disease risk prediction, individualized nutritional intervention, and precision medicine, with broad clinical application prospects, but larger-scale multicenter studies are still needed to validate its association strength and specific molecular mechanisms across different populations.
