Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.

3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.

22Rv1 is an adherent, epithelial-like human prostate carcinoma cell line established from a serially transplanted CWR22R xenograft. The parental cells retain androgen receptor signaling and express prostate-specific antigen, while also carrying constitutively active androgen receptor splice variants, including AR-V7. These features have made 22Rv1 a widely used model for studying castration-resistant prostate cancer and resistance to androgen receptor–directed therapies. Nluc Reporter Cell Line - 22Rv1 is an engineered derivative that expresses NanoLuc luciferase, enabling cellular activity to be quantified through a sensitive bioluminescent readout. NanoLuc is a compact 19.1 kDa luciferase that produces bright, ATP-independent luminescence in the presence of a compatible furimazine-based substrate. The reporter format therefore offers a practical means of detecting and following 22Rv1 cells without relying solely on endpoint staining or cell counting.

This reporter cell line is suited to prostate cancer studies in which a broad and easily measurable signal is needed. In vitro, NanoLuc activity can support the assessment of cell proliferation, survival, cytotoxicity, and treatment response across multiwell assay formats. The 22Rv1 background is particularly relevant to investigations of androgen receptor signaling, AR-V7-associated drug resistance, and responses to antiandrogens or pathway-targeted compounds. The cells may also be used to evaluate candidate small molecules, biologics, immune-cell therapies, and combination regimens, provided that appropriate assay controls are included. In validated animal models, bioluminescence can facilitate longitudinal monitoring of tumor establishment and changes in tumor burden, reducing dependence on repeated terminal measurements. This combination of a clinically relevant prostate cancer background and a sensitive reporter readout makes the cell line useful for compound screening, mechanism-of-action studies, and preclinical efficacy research.
