The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

The RENCA cell line is a highly characterized murine model of renal cell carcinoma, originally derived from a spontaneous cortical renal cell tumor isolated from a BALB/c mouse. It serves as a cornerstone in urological oncology research, particularly valued as a syngeneic model that faithfully replicates the complex tumor microenvironment, aggressive local growth, and metastatic progression characteristic of advanced human kidney cancer. The mScarlet/Luc Reporter Cell Line - RENCA is a sophisticated, stably transfected derivative explicitly engineered to simultaneously co-express both the exceptionally bright, monomeric red fluorescent protein mScarlet and firefly luciferase (Luc). This advanced dual-labeling construct ensures continuous, high-level expression of both functional reporter genes across sequential in vitro passages. Crucially, this stable genetic integration is achieved without compromising the fundamental epithelial morphology, the intrinsic biological properties, or the highly tumorigenic and metastatic capacity of the original parental RENCA cells.
The incorporation of this dual-reporter system significantly broadens the experimental utility of the RENCA model for comprehensive translational renal cancer research. For in vitro studies, the intense red fluorescence provided by mScarlet expression allows researchers to easily visualize cellular dynamics, accurately quantify proliferation rates, and seamlessly isolate specific cellular subpopulations using standard fluorescence microscopy and flow cytometry techniques. For in vivo applications, the robust luciferase activity serves as the optimal foundation for non-invasive, longitudinal tracking of primary tumor progression and distant metastatic dissemination (frequently to the lungs and liver) through bioluminescence imaging (BLI). Because it is a murine-derived cell line, the RENCA-mScarlet/Luc model is engrafted into immunocompetent, syngeneic BALB/c mice to establish clinically relevant orthotopic subcapsular renal models or robust models of experimental metastasis. This specialized reporter line represents an exceptionally valuable platform for evaluating the in vivo therapeutic efficacy of novel anti-angiogenic compounds, targeted pharmacological agents, and—crucially—emerging immunotherapies (such as immune checkpoint inhibitors), facilitating the real-time, accurate quantification of localized tumor burden and disseminated disease in living subjects with fully intact immune systems.

We used the RENCA mScarlet/Luc cell line from Creative Biogene for our immunocompetent mouse models. The mScarlet fluorescence provided excellent contrast against tissue autofluorescence, and the luciferase activity was highly stable throughout the 5-week study. Great quality and it saved us months of lentiviral development!
