Safety Considerations

The following safety precautions should be observed.

1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.

2. No eating, drinking or smoking while handling the stable line.

3. Wash hands after handling the stable line and before leaving the lab.

4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.

5. All waste should be considered hazardous.

6. Dispose of all liquid waste after each experiment and treat with bleach.

The Otof gene encodes otoferlin, a multi-C2 domain transmembrane protein crucial for hearing and balance in vertebrates. Otoferlin acts as a primary calcium ion sensor for exocytosis at the inner hair cell (IHC) synaptic band, responsible for the rapid release of neurotransmitters. Clinically, biallelic mutations in the Otof gene lead to DFNB9 congenital deafness. OTOF-targeted gene therapy using adeno-associated virus (AAV) vectors has shown promising results in clinical trials. Otoferlin is also involved in vesicle circulation and the maintenance of synaptic structure.

The stable mouse Otof cell line in the HEK293T context provides an ideal platform for evaluating the function of otoferlin and its gene therapy constructs. The HEK293T cell line is well-suited for expressing large proteins and producing high-titer AAV vectors. By stably overexpressing mouse Otof, this cell line provides a standardized environment for studying calcium ion sensing properties and their interactions with membrane transport proteins such as SNARE proteins. This product has invaluable value for screening compounds that regulate hair cell exocytosis and assessing the safety of AAV-OTOF.
