Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

L929, also known as NCTC clone 929 or L-929, is an adherent mouse fibroblast-like cell line derived from subcutaneous areolar and adipose tissue of a male C3H/An mouse. Established in 1948 from the parental strain L, it was among the earliest continuously cultured clonal mammalian cell lines. L929 cells are widely used in cytotoxicity testing and are particularly well known for their sensitivity to tumor necrosis factor alpha–induced cell death. GFP/Luc Reporter Cell Line - L929 is an engineered derivative expressing green fluorescent protein and luciferase. GFP permits direct visualization and identification of labeled cells, while luciferase produces a sensitive bioluminescent signal after addition of an appropriate substrate. The two reporters provide complementary options for imaging and quantitative assessment of the same cell population.

This reporter line can support cell viability, proliferation, cytotoxicity, and cell-tracking studies. GFP fluorescence is useful for microscopy, flow cytometry, and analysis of cell morphology or spatial distribution in co-culture and three-dimensional models. Luciferase activity offers a convenient quantitative readout for multiwell assays, including dose–response experiments and screening of compounds or biomaterials. Given the established use of parental L929 cells in TNF bioassays, the reporter derivative may also assist studies of TNF-mediated apoptosis and necroptosis, provided that reporter expression has not altered the expected cytokine response. In compatible animal models, bioluminescence may be used to follow the localization, persistence, or expansion of implanted cells over time, while GFP can support subsequent ex vivo examination of tissues.
