Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. The C666-1 cell line is a well-characterized, EBV-positive undifferentiated nasopharyngeal carcinoma (NPC) line originally established from a Southern Chinese patient. It remains one of the few NPC lines that consistently maintains the Epstein-Barr virus genome in culture, making it a standard model for studying EBV-driven carcinogenesis. The C666-1-GFP/Luc reporter line was generated by stably transfecting parental C666-1 cells with a dual-expression construct encoding enhanced green fluorescent protein (eGFP) and firefly luciferase under a constitutive promoter. This enables simultaneous fluorescence-based tracking and bioluminescent quantification of viable cells. The line is typically selected with puromycin and validated for stable reporter expression, mycoplasma negativity, and retained EBV status.
This dual-reporter system supports a range of experimental applications. The luciferase signal allows non-invasive longitudinal monitoring of tumor burden in xenograft models using bioluminescence imaging, which is particularly useful for assessing therapeutic responses or metastatic spread.
