Glypican-1 (GPC1) is a cell surface heparan sulfate proteoglycan encoded by the GPC1 gene located on human chromosome 2q37.3. The protein consists of 558 amino acids and is anchored to the plasma membrane via a glycosylphosphatidylinositol (GPI) linkage, with three heparan sulfate chains attached to its core. GPC1 plays important roles in regulating cell division, growth factor signaling, and axon guidance by interacting with molecules such as fibroblast growth factors, VEGF, and SLIT2. The Human GPC1 Stable Cell Line-HEK293 is generated by transfecting the human GPC1 gene into HEK293 cells, a widely used mammalian expression system known for its high transfection efficiency and reliable protein expression. These cells stably express GPC1 on the cell surface, providing a consistent and reproducible in vitro model for studying the biological functions of this proteoglycan.
This stable cell line serves as a valuable tool for multiple research applications. It can be used to investigate GPC1-mediated signaling pathways, including its role in modulating fibroblast growth factor receptor signaling and Wnt pathway activity. Researchers employ this cell line for antibody discovery and validation, particularly for developing therapeutic antibodies targeting GPC1, which has emerged as a promising candidate for cancer therapy. GPC1 is overexpressed in several malignancies including pancreatic cancer, breast cancer, and prostate cancer, making this cell line useful for studying tumor biology and evaluating potential therapeutics such as antibody-drug conjugates, CAR-T cells, and immunotoxins. The line also supports functional studies on GPC1 interactions with growth factors and extracellular matrix components, as well as screening assays for small molecule inhibitors or biologics that target GPC1.
