The CLDN4 gene, located on human chromosome 7q11.23, encodes claudin-4, a member of the claudin family of integral membrane proteins. Claudin-4 is a core structural component of epithelial tight junctions, where it plays a key role in regulating the paracellular transport of ions and solutes across epithelial barriers. This intronless gene also codes for a high-affinity receptor for Clostridium perfringens enterotoxin (CPE), and its deletion is associated with Williams-Beuren syndrome, a multisystem neurodevelopmental disorder. The Human CLDN4 Stable Cell Line-HEK293 is generated by stably transfecting the human CLDN4 gene into HEK293 cells, a widely used human embryonic kidney cell line valued for its high transfection efficiency, reliable protein expression, and scalability. This stable cell line provides a consistent and reproducible in vitro model for studying claudin-4 biology, as it constitutively expresses the target protein at elevated levels on the cell surface.
This cell line serves as a versatile tool for a range of research applications. It is particularly useful for compound screening and functional studies targeting claudin-4, given its well-documented overexpression in multiple cancer types including prostate, ovarian, breast, pancreatic, and bladder carcinomas. Researchers can employ this line to evaluate the efficacy of therapeutic antibodies, small molecules, or CPE-based cytotoxic agents that rely on claudin-4 binding for tumor cell targeting. Additionally, it supports investigations into tight junction modulation, epithelial barrier function, and paracellular permeability, making it applicable to drug transport studies and gastrointestinal or renal physiology research. The line also facilitates mechanistic studies on claudin-4 interactions with scaffolding proteins such as TJP1, and its downstream signaling via pathways like AKT and ERK1/2.
