Claudin-18.2 (CLDN18.2) is a promising target for immunotherapy, particularly in gastrointestinal tumors such as gastric cancer. Its expression has been linked to advanced cancer stages, poor prognosis, and increased metastasis. The researchers investigated the role of CLDN18.2 in gastric cancer cell adhesion to cancer-associated fibroblasts (CAFs) using in vitro and in vivo models. They found that CLDN18.2 facilitates the interaction between cancer cells and CAFs, contributing to tumor progression and metastasis. Additionally, RNA sequencing and protein proximity labeling revealed that S100A4, a CAF marker, interacts with CLDN18.2 to promote gastric cancer progression.
Figure 1. The researchers utilized paraffin immunofluorescence to analyze PDX tumors with different CLDN18.2 staining levels. They applied antibodies against CLDN18.2, FAP, α-SMA, and S100A4 to assess the expression and infiltration of CAFs. (Liu S, et al., 2024)
Creative Biogene's Human CLDN18-A2 Stable Cell Line - HEK293T can be a valuable tool for further research on CLDN18.2-related mechanisms. This cell line enables the study of CLDN18.2 interactions and its role in cancer progression, particularly in gastric cancer models.
