AHCYL1, also known as adenosylhomocysteinase-like 1, is a protein-coding gene in humans that encodes an enzyme with structure and function similar to adenosylhomocysteinase. The gene is located at position 13.3 of the short arm of chromosome 1 and is approximately 39 kilobases in length. The gene is expressed in various tissues including the brain, particularly in areas such as the globus pallidus, ventral tegmental area, and supravesicular nucleus. The AHCYL1 gene is involved in many important biological processes. It plays a crucial role in the S-adenosylmethionine cycle, which is essential for cellular metabolism and methylation reactions. This cycle is essential for regulating various physiological processes, including the methylation of DNA, RNA, proteins, and lipids. AHCYL1 is known to interact with inositol 1,4,5-triphosphate (IP3) receptors, specifically ITPR1. This interaction is important for various cellular processes, including the calcium signaling pathway. Furthermore, AHCYL1 activity has been implicated in the regulation of mRNA polyadenylation and positive regulation of sodium ion transport, further highlighting its broad functional impact in cellular homeostasis.
The following are the main applications of the Human AHCYL1 Knockout Cell Line-HEK293: Gene Function Analysis: The AHCYL1 knockout cell line allows researchers to explore the functional role of the AHCYL1 gene in cellular processes. Disease Modeling: This cell line can be used to model diseases associated with mutations or dysregulation of the AHCYL1 gene. This helps understand the mechanisms underlying such diseases and aids in the development of targeted therapies and interventions. Drug Screening: Researchers can use AHCYL1 knockout HEK293 cells to screen for drugs that target pathways affected by AHCYL1. This provides a platform for identifying potential therapeutic compounds and understanding their mechanisms of action. Signal Transduction Studies: The AHCYL1 gene is involved in signaling pathways within cells. Knockout cell lines help reveal the role of AHCYL1 in these pathways and can lead to discoveries about cellular signaling mechanisms and their impact on health and disease. Protein-Protein Interactions: The absence of AHCYL1 in these cells allows for the study of protein-protein interactions and helps identify interacting partners of AHCYL1.
Customer Q&As
What is the recommended growth medium? Does it require antibiotic selection?

A: DMEM supplemented with 10% fetal bovine serum.
It is not required to add the selection antibiotics when culturing the KO cells.

How is the knockout cell line validated?

A: The knockout cell product is validated by PCR amplification and Sanger Sequencing to confirm the mutation at the genomic level. Please find the detailed mutation info in the datasheet.

Is the product a single clonal cell or mixed cell pool?

A: Single clonal cell.

Can I confirm gene knockout by RT-qPCR?

A: No. This knockout cell product is generated using the CRISPR/Cas9 system to induce small insertions or deletions (indels) resulting in frameshift mutations. Although these frameshift mutations typically disrupt the coding gene, there is a possibility that the non-functional transcript may still be transcribed. Consequently, this could potentially yield misleading results when analyzed by RT-qPCR.

How can I store the cell product?

A: The cell line should be stored in liquid nitrogen for long-term preservation.

Is it possible to get multiple knockout clones for my GOI?

A: For most cases, we often keep at least 2 clones with different frameshift mutations. Please feel free to contact us to check if there are additional available clones.
