The TGF-β2 protein, encoded by the TGFB2 gene, belongs to the TGF-β family. Although TGF-β ligands and their corresponding receptors are expressed in most human tissues, tumor cells remain a significant source of TGF-β production. Secreted TGF-β binds to its receptors and initiates a signaling cascade within the nucleus via cytoplasmic signal-mediating molecules (Smads). The TGF-β/Smad complex regulates the expression of downstream target genes, thereby modulating cellular biological functions. In malignant tumor cells, the TGF-β/Smad pathway is frequently mutated, and the level of TGF-β overexpression correlates with tumor malignancy. TGF-β influences breast cancer (BC) progression by inducing anti-apoptotic effects, promoting immune escape, and facilitating cell proliferation and migration in BC cells. Here, the researchers demonstrated that TGFB2 overexpression reverses the tumor-suppressive effects of M511-0965, indicating that TGFB2 is a key downstream effector gene of M511-0965.

Here, the researchers generated TGFB2-overexpressing TNBC cells (Figure 1A). Western blot analysis revealed that in TGFB2-overexpressing cells, BAX levels were reduced, BCL2 expression was upregulated, and apoptosis was inhibited. Furthermore, TGFB2 overexpression promoted the epithelial-mesenchymal transition (EMT) and accelerated metastasis in TNBC cells. Importantly, M511-0965 reversed the effects of TGFB2 overexpression on the inhibition of apoptosis and the promotion of metastasis (Figure 1B). Functional assays confirmed that M511-0965 counteracted the promoting effects of TGFB2 overexpression on cell proliferation, invasion, and migration (Figures 1C-I).
