Overactivation of AKT drives the malignant phenotype of lung cancer by promoting tumor cell proliferation and survival. However, the relationship between dysregulated cell cycle progression and AKT1 kinase activity remains unclear. Here, researchers found that PKMYT1 is downregulated in lung adenocarcinoma (LUAD) tissues, and its low expression predicts poor prognosis in LUAD patients. PKMYT1 exerts a potent tumor-suppressive function in LUAD cells by inhibiting AKT1 activation and subsequently suppressing cell cycle progression, which depends on its tyrosine and threonine protein kinase activity. Interestingly, PKMYT1 can directly bind to AKT1, thereby inhibiting AKT1 activation. Furthermore, silencing AKT1 and using inhibitors targeting the AKT pathway effectively reversed the promoting effects of PKMYT1 knockdown on LUAD cell proliferation, migration, and invasion. These studies reveal the antitumor role of PKMYT1 in LUAD and provide evidence for elucidating the dual role of PKMYT1 in tumor progression.

Here, researchers investigated the function of PKMYT1 in lung adenocarcinoma (LUAD) cells. Overexpression of PKMYT1 significantly slowed the proliferation rate of A549 and H1299 cells, which have low basal expression levels of PKMYT1 (Figure 1a and 1b). Compared to the control group, PKMYT1-overexpressing A549 and H1299 cells formed fewer and smaller colonies in culture dishes and soft agar medium (Figure 1c and 1d). Furthermore, compared to control cells, PKMYT1-overexpressing A549 and H1299 cells showed reduced migration and invasion capabilities, with significantly fewer cells passing through the Transwell chambers and wider wound healing gaps (Figure 1e). Conversely, in PKMYT1-knockdown BEAS-2B cells, cell viability and migration ability were enhanced (Figure 1a-c and 1e). Simultaneously, silencing PKMYT1 promoted the proliferation (Figure 1a-c), anchorage-independent growth (Figure 1d), migration, and invasion (Figure 1e) of H1975 cells. In summary, PKMYT1 inhibits the malignant phenotype of LUAD cells in vitro.
