| Summary | Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance. |
| Sterility | Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination. |
| Ad5 E1 Detection | All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination. |
| RCA Assays | Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources. |
| PFU Titering | All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells. |

The NAB2 (NGFI-A binding protein 2) gene encodes a transcriptional repressor that plays a key role in regulating cell growth and differentiation. NAB2 is located on human chromosome 12 and regulates its activity by binding to the early growth response (EGR) family of transcription factors, particularly EGR1. This interaction is crucial for controlling processes such as cell proliferation, apoptosis, and wound healing. Dysregulation of NAB2 is associated with multiple pathological conditions, including cancer and fibrosis. Notably, chromosomal rearrangements involving NAB2 and STAT6 are frequently observed in solitary fibrous tumors (SFTs), resulting in the formation of NAB2-STAT6 fusion proteins that drive oncogenic signaling.
The NAB2 adenovirus is a recombinant adenoviral vector used to deliver or manipulate the NAB2 gene for research or therapeutic purposes. Adenoviruses are widely used in gene therapy due to their high transduction efficiency, ability to infect both dividing and non-dividing cells, and relatively large cargo capacity. The NAB2 adenovirus can be used to overexpress NAB2 to study its inhibitory effects on EGR-mediated transcription. Such vectors are valuable tools for exploring the role of NAB2 in cancer biology, fibrosis, and other diseases associated with aberrant EGR signaling. Furthermore, NAB2 adenoviruses may have therapeutic potential, particularly in cancers driven by NAB2-STAT6 fusions, where targeted gene editing or silencing could inhibit tumor growth.
