Description
Knockout Detection Kit provides a simple and highly effective method to detect gene knockout and gene polymorphism. The core component of the kit is the knockout detection enzyme, which has strong endonuclease activity and is homologous to Cel. The knockout detection enzyme can efficiently recognize and cleave the mismatches in heterogeneous DNA duplexes. Our knockout detection kit can be widely used for gene knockout detection in mammals, bacteria, fungi, virus and plants, especially applicable to screen positive clones and detect knockout efficiency using CRISPR/Cas9, TALENs or ZENs technologies.
Applications
The Knockout Detection Kit can be applied for knockout detecting of many kinds of nucleases, such as CRISPR/Cas9, TALEN and ZFN and so on.
Efficiency
5-20 min of digestion time is enough to detect any type of mutations such as substitution, insertion and deletion.

How is gene knockout detected?

A: When knocking out a gene, the levels of protein expression should be altered and thus measurements of protein expression can also be used to validate a successful CRISPR knockout further. This can be accomplished by the Western Blot technique or by mass spectrometry.

Is gene silencing the same as knockout?

A: No. Gene silencing is considered a gene knockdown mechanism, which usually reduces the expression of a gene but does not completely eliminate it. In contrast, gene knockout completely erases genes from the organism's genome; therefore, there's no expression of those genes at all.

What is the difference between transgenic and knockout?

A: Two important tools used by researchers include transgenic mice, in which a foreign gene is integrated into an animal's genetic material, while in knock-out/knock-in mice, the target genes either lose function or are changed.
