Nemo-like kinase (NLK), encoded by the human NLK gene, is a serine/threonine protein kinase of the mitogen-activated protein kinase (MAPK)-like family. In mammals, NLK plays a key regulatory role in multiple fundamental signal transduction cascades, most notably the Wnt/β-catenin, transforming growth factor-β (TGF-β), and Notch pathways. It acts downstream of a variety of stimuli, typically by directly phosphorylating key transcription factors such as TCF/LEF (in Wnt signaling) and STAT3, thereby regulating their transcriptional activity, subcellular localization, or stability. This phosphorylation often results in the inhibition of target gene expression. Therefore, NLK plays an integral role in a variety of cellular processes including cell proliferation, differentiation, apoptosis, migration, and neurogenesis. Dysregulation of NLK expression or activity has been implicated in a variety of pathological conditions, including several cancers (where it can act as a tumor suppressor or promoter, depending on the context), neurological diseases, and developmental defects.
