The neuronal guanine nucleotide exchange factor (NGEF) gene, also known as Ephexin-1, is located on chromosome 2q31.1 in humans. It encodes a key Rho guanine nucleotide exchange factor (RhoGEF) protein that is primarily expressed in the nervous system. NGEF acts as a molecular switch activator that specifically catalyzes the exchange of GDP for GTP on RhoA and RhoC GTPases, key regulators of the actin cytoskeleton. This function places NGEF at the heart of key cellular processes, including neuronal development, axon guidance, dendritic spine morphogenesis, and synapse formation. Its activity influences neuronal migration, growth cone dynamics, and ultimately the complex connectivity of the brain. Beyond neurobiology, NGEF expression has been detected in other tissues and has been associated with a variety of cancers.
Human NGEF adenoviral particles are sophisticated, replication-defective viral vectors engineered to efficiently deliver human NGEF gene sequences to a variety of mammalian cell types in vivo and in vitro. These particles are based on a modified human adenovirus serotype 5 (Ad5) in which essential viral replication genes (usually the E1 region, and often the E3 region) have been deleted. This deletion renders the virus incapable of independent replication in standard host cells, significantly improving biosafety for research applications. Researchers have used human NGEF adenoviral particles to study its function by overexpressing the gene in target cells or model organisms, investigating its effects on Rho GTPase signaling, cytoskeletal dynamics, neuronal morphology and function, cell migration, and its role in cancer progression.
