Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance.
Sterility Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination.
Ad5 E1 Detection All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination.
RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.

The CLDN2 gene encodes the tight junction protein Claudin-2, an essential component of tight junctions that regulate paracellular permeability and maintain the integrity of epithelial barriers in a variety of tissues, including the kidney, liver, and intestine. Claudin-2 selectively forms pores for cations and small molecules, playing a key role in ion homeostasis, cell adhesion, and tissue-specific physiological functions. Dysregulated CLDN2 expression has been implicated in a variety of pathological conditions, such as chronic kidney disease, liver fibrosis, and inflammatory bowel disease (IBD), where barrier dysfunction exacerbates disease progression. In addition, CLDN2 overexpression has been associated with cancer metastasis, particularly in gastrointestinal malignancies. Due to its involvement in these processes, CLDN2 has become an important target for research in epithelial biology, drug delivery mechanisms, and therapeutic interventions.

Human CLDN2 Adenoviral Particles are high-titer, replication-defective viral vectors designed to deliver the CLDN2 gene to mammalian cells for functional studies or therapeutic applications. These particles take advantage of the high transduction efficiency, broad tropism, and ability of adenovirus to infect both dividing and non-dividing cells, making them ideal for in vitro and in vivo studies. The engineered adenoviral backbone ensures stable expression of CLDN2 under the control of a strong promoter, allowing researchers to study the role of this gene in epithelial barrier regulation, disease mechanisms, or potential gene therapy strategies. Prepackaged adenoviral particles are purified and ready for use, reducing experimental variability and saving time compared to traditional transfection methods. Applications include studying the effect of CLDN2 on tight junction dynamics, modeling diseases such as inflammatory bowel disease (IBD) or fibrosis, or screening compounds targeting claudin-2-related pathways.
