The Bruton''s tyrosine kinase (BTK) gene is located on chromosome X (Xq22.1) and encodes a critically important cytoplasmic non-receptor tyrosine kinase belonging to the Tec kinase family. BTK plays a key role in signal transduction pathways and is primarily found in hematopoietic cells, especially B lymphocytes and myeloid cells. Its expression begins early in B-cell development in the bone marrow and persists throughout most stages of B-cell maturation (except plasma cells). Upon activation by upstream signals, usually initiated by the binding of the B-cell receptor (BCR) or certain cytokine receptors, BTK undergoes phosphorylation and membrane recruitment. It then functions as a key amplifier in the BCR signaling cascade, culminating in the activation of phospholipase Cγ2 (PLCγ2), calcium mobilization, and downstream pathways that regulate B-cell proliferation, differentiation, survival, and antigen presentation. Therefore, mutations in the BTK gene, especially loss of function, lead to X-linked agammaglobulinemia (XLA), a primary immunodeficiency disorder characterized by a severe lack of mature B cells and antibodies. In contrast, BTK is constitutively active and is a key driver of certain B-cell malignancies, particularly chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), making it a major therapeutic target.
Human BTK adenoviral particles are a powerful and widely used recombinant viral vector system specifically designed to deliver the human BTK gene to a variety of mammalian cells (in vitro and in vivo). Human BTK adenoviral particles are widely used to: 1) restore BTK function in BTK-deficient cell lines or primary cells (e.g., cells from XLA patients) to study signaling pathways and rescue phenotypes; 2) overexpress wild-type or mutant BTK in cell lines or primary cells to study its role in signal transduction, cell survival, proliferation, migration, and tumorigenesis; 3) screen BTK inhibitors by establishing cell models with defined BTK expression levels; 4) construct cell models of diseases such as chronic lymphocytic leukemia (CLL) or evaluate the role of BTK in other immune cells; 5) develop gene therapy strategies.
