The AGER gene (receptor for advanced glycosylation end products) is located on human chromosome 6p21.3 and encodes the receptor for advanced glycosylation end products (RAGE). RAGE is a multi-ligand pattern recognition receptor that belongs to the immunoglobulin superfamily. Its name derives from its initial identification as a receptor binding advanced glycosylation end products (AGEs). It is a key signaling receptor for multiple damage-associated molecular patterns (DAMPs). Upon ligand binding, RAGE typically activates central pro-inflammatory pathways such as NF-κB and MAP kinases (e.g., p38, JNK), resulting in the production of cytokines, chemokines, and adhesion molecules. It also promotes oxidative stress by activating NADPH oxidase. Therefore, RAGE is closely associated with the pathogenesis and progression of many chronic diseases, including diabetes and its complications (retinopathy, nephropathy, neuropathy, atherosclerosis), Alzheimer''s disease, inflammatory diseases (arthritis, colitis), cancer progression and metastasis, and chronic obstructive pulmonary disease (COPD). Its expression is usually low in healthy adult tissues, but is significantly upregulated at sites of inflammation, injury, and pathology, making it an important therapeutic target.

Human AGER adenoviral particles are an advanced molecular biology tool specifically designed to deliver and express the human AGER gene into a variety of mammalian cell types in vitro or in vivo. These particles are based on an engineered replication-defective adenovirus serotype 5 (Ad5) vector. Its core genetic component is a recombinant adenoviral genome in which essential viral replication genes (E1 region and usually E3 region) have been deleted.
