A: TOPO cloning is a molecular biology technique in which DNA fragments are cloned into specific vectors without the requirement for DNA ligases.
A: DNA topoisomerase enzyme has sequence-specific DNA cleavage and ligation activity. In TOPO TA cloning, the T overhang containing vector ends are attached with the TOPO enzyme. The topoisomerase enzyme seals the joints during the stable association of the vector and insert ends using AT base-pairing.
