| Summary | Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance. |
| Sterility | Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination. |
| Ad5 E1 Detection | All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination. |
| RCA Assays | Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources. |
| PFU Titering | All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells. |

CPA3, or carboxypeptidase A3, belongs to the carboxypeptidase family, a class of zinc-dependent metalloproteinases. The CPA3 gene is located on human chromosome 7 and is primarily expressed in mast cells and certain organs, such as the pancreas. This gene plays a crucial role in a variety of biological processes, including protein processing, inflammation regulation, and neuropeptide activity. The CPA3 enzyme specifically cleaves basic amino acids from the carboxyl terminus of peptides, thereby affecting the activity of peptide hormones and participating in the inactivation of bioactive peptides. Aberrant expression or mutations in the CPA3 gene are associated with a variety of diseases, including allergic reactions and certain types of cancer.
The CPA3 adenovirus is a recombinant viral vector genetically engineered to selectively target and infect cells expressing the CPA3 protein. Adenovirus offers numerous advantages as a gene therapy vector, including efficient delivery of therapeutic genes to target cells, high transduction efficiency, and the potential to elicit a potent immune response against tumor cells. The CPA3 adenovirus can be used to deliver therapeutic genes to induce apoptosis (programmed cell death) in cancer cells expressing CPA3, or provide a way to specifically introduce suicide genes in the tumor microenvironment to convert harmless prodrugs into toxic substances.
