The CBFA2T1 (core binding factor, runt domain, alpha subunit 2, translocation to 1) gene, also known as ETO or MTG8, is a key transcriptional regulator involved in hematopoiesis, cell differentiation, and tumorigenesis. CBFA2T1 is located on chromosome 8q21 and encodes a nuclear protein that functions as a transcriptional corepressor by interacting with histone deacetylases (HDACs) and other chromatin-modifying complexes. It plays a key role in repressing target genes associated with cell proliferation and survival. Notably, CBFA2T1 is frequently associated with chromosomal translocations, such as the t(8;21) translocation in acute myeloid leukemia (AML), where it fuses with the RUNX1 gene and disrupts normal hematopoietic differentiation.
