The BAP1 (BRCA1-associated protein 1) gene is a tumor suppressor gene located on chromosome 3p21.1. It encodes a nuclear ubiquitin carboxyl-terminal hydrolase that plays a key role in regulating cellular processes such as DNA damage repair, cell cycle regulation, and chromatin remodeling. BAP1 acts as a deubiquitinating enzyme that regulates the activity of key proteins involved in proliferation and apoptosis. Germline mutations in BAP1 are associated with an increased risk of developing a variety of cancers, including mesothelioma, uveal melanoma, renal cell carcinoma, and other malignancies, a condition known as the BAP1 tumor predisposition syndrome. Somatic mutations or loss of BAP1 expression are also common in sporadic cancers. Due to its involvement in key signaling pathways, BAP1 has emerged as a promising therapeutic target for cancer treatment.
