Summary Creative Biogene ensures high-quality AAV particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between AAV particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in the production of AAV, especially for applications in animal studies and gene therapy. Effective endotoxin quality control is essential in the development and manufacturing of AAV particles. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced AAV particles to ensure regulatory compliance.
Purity AAV purity is critical for ensuring the safety and efficacy of AAV-based applications.AAV capsids are composed of three main protein components, known as viral proteins: VP1, VP2, and VP3. These proteins play a critical role in the structure and functionality of the AAV capsid. Monitoring the VP1, VP2, and VP3 content in AAV preparations is essential for quality control in AAV production. Our AAV particles are tested for showing three clear bands of VP1, VP2 VP3 by SDS-PAGE.
Sterility The AAV virus samples are inoculated into the cell culture medium for about 5 days to detect bacterial and fungal growth.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of AAV to deliver genetic material into target cells or tissues, and assess gene expression and functional activities.
Empty vs. Full Capsids Based-on our proprietary AAV production and purification technology, Creative Biogene can always offer AAV particles with high ratio of full capsids. If required, we can also assess the ratio for a specifc lot of AAV particles by transmission electron microscopy (TEM) or other methods.

Recombinant adeno-associated viral vectors (AAV) have an excellent safety profile and do not cause any known human or animal disease. Therefore, they are considered one of the preferred vectors for clinical applications. AAV has several advantages, including the ability to infect both dividing and quiescent cells, and the rarity of host genome integration reduces the risk of insertional mutagenesis. AAV-mediated transgene expression is not permanent but can persist for months to years, depending on the immune response and the turnover rate of infected cells. Therefore, inactivation of the transgene may not be necessary after the disease is cured. Finally, AAV is significantly less immunogenic than adenoviral vectors, but because AAV is widely present in nature, more than 70% of the population is seropositive to one or more AAV serotypes.

New chimeric capsids have been created based on 12 known natural serotypes to further reduce immunogenicity. The AAV-DJ serotype is a chimera of the AAV2/8/9 serotypes, differing from its closest natural relative (AAV-2) by 60 capsid amino acids. An important feature of the AAV-DJ capsid is that it was artificially selected using pooled human antisera. As a result, the capsid has significantly increased resistance to AAV neutralizing antibodies.

Episodic memories are stored in distributed neurons, but how individual neurons are determined to be fit for a coding ensemble remains unclear. Here, researchers identified thousands of regions that are predominantly bistable (CpG methylated or unmethylated) within neuronal genomes established during development in the mouse hippocampal dentate gyrus. Reducing DNA methylation and the proportion of methylated epitopes in bistable regions impairs novel context-induced neuronal activation and spatial memory. Conversely, increasing the frequency of methylated and unmethylated epitopes in bistable regions enhances intrinsic excitability and spatial memory but impairs spatial working memory, suggesting that the methylated-unmethylated epitope balance established during development in bistable regions is critical for normal neuronal excitability and hippocampal cognitive function. Researchers propose a model in which epigenetic bistable regions generate neuronal heterogeneity and that specific combinations of exonic epialleles determine the fitness of the coding ensemble by modulating neuronal excitability.

Adult mice were injected bilaterally in the dDG with either DNMT3A expressing AAV-DJ-SYN-DNMT3a-GFP virus or control AAV-DJ-SYN-GFP virus, titrated to achieve a relatively sparse expression pattern (Figure 1a). Twenty days later, animals injected with both Dnmt3a and control viruses exhibited GFP expression in the DGC. Overexpression of DNMT3A (OE) resulted in population-level hypermethylation of a large number of CpG sites, albeit restricted to a small subset of cells. These CpGs, like IMR CpGs, showed predominantly IM in control cells (Figure 1b, pink), but were relatively hypermethylated in OE neurons, showing a rightward shift in methylation (Figure 1b, blue). Unaffected CpGs in OE cells exhibited a bimodal distribution, with no clear rightward shift in their methylation profile (Figure 1c), indicating that hypermethylation in OE cells is primarily restricted to IM CpGs. Notably, the hypermethylated CpGs clustered into 13,745 DMRs (>2 CpGs), which largely overlapped with previously identified IMRs (67.38%, Fig. d). Furthermore, most genes associated with OE-DMRs were also IMR genes (Figure 1e). Similar to the hypomethylation in cKO, the hypermethylation in OE was partial, as not all IMR genes were affected and only some of the IMRs in genes with multiple IMRs were hypermethylated. Overall, these data indicate that IMRs can be hypermethylated, at least partially, by the OE of Dnmt3a.

Overexpression of DNMT3A in DGCs increases methylation at a subset of IMRs.Figure 1. Overexpression of DNMT3A in DGCs increases methylation at a subset of IMRs. (Odell S C, et al., 2020)
