| Summary | Creative Biogene ensures high-quality AAV particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between AAV particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in the production of AAV, especially for applications in animal studies and gene therapy. Effective endotoxin quality control is essential in the development and manufacturing of AAV particles. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced AAV particles to ensure regulatory compliance. |
| Purity | AAV purity is critical for ensuring the safety and efficacy of AAV-based applications.AAV capsids are composed of three main protein components, known as viral proteins: VP1, VP2, and VP3. These proteins play a critical role in the structure and functionality of the AAV capsid. Monitoring the VP1, VP2, and VP3 content in AAV preparations is essential for quality control in AAV production. Our AAV particles are tested for showing three clear bands of VP1, VP2 VP3 by SDS-PAGE. |
| Sterility | The AAV virus samples are inoculated into the cell culture medium for about 5 days to detect bacterial and fungal growth. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of AAV to deliver genetic material into target cells or tissues, and assess gene expression and functional activities. |
| Empty vs. Full Capsids | Based-on our proprietary AAV production and purification technology, Creative Biogene can always offer AAV particles with high ratio of full capsids. If required, we can also assess the ratio for a specifc lot of AAV particles by transmission electron microscopy (TEM) or other methods. |
This AAV-DJ chimera was engineered to have enhanced liver tropism and resistance to pre-existing NAbs and was made using a two-pronged approach. First, capsid genes from eight different AAVs (AAV2, AAV4, AAV5, AAV8, and AAV9, as well as animal isolates AAAV, BAAV, and CAAV from avian, bovine, and caprine species, respectively) were combined to create a library of shuffled AAV capsids. This library was sequentially screened on human hepatoma cells (Huh-7 and HepG2), which have a low natural tolerance to AAV infection. The resulting clones were further selected in the presence of pooled human antisera (intravenous immunoglobulins) that are neutralizing against many AAV serotypes and then amplified on hepatoma cells for multiple rounds of directed evolution.
AAV-DJ chimeras, which contain capsid VP contributions from AAV2, AAV8, and AAV9, have enhanced in vivo liver tropism, the ability to evade known NAbs, and efficiently deliver the factor IX transgene to liver tissue in both naive mice and mice treated with intravenous immunoglobulin. By cryo-reconstruction, the structure of AAV-DJ was shown to retain the HS receptor attachment site of the parental AAV2, while the presence of structural differences in VR-I (contributed by AAV9), a loop known to be involved in binding the A20 NAb. Thus, cellular recognition is maintained, and this variation explains the ability of the chimera to evade recognition by A20 and potentially other NAbs that bind at VR-I.
Adeno-associated virus (AAV) vectors are widely used to treat inherited retinal diseases, but their transduction efficiency by intravitreal injection still needs to be improved. Here, the researchers explored the transduction efficiency of AAV-DJ(K137R)-GFP in different retinal cells of normal mice and the therapeutic effect of AAV-DJ(K137R)-Rs1 on the retinal function and structure of Rs1-KO mice. Intravitreal injection of AAV-DJ(K137R)-GFP showed that the vector transduced cells in various layers of the retina, including the inner nuclear layer and the photoreceptor layer. Intravitreal injection of AAV-DJ(K137R)-Rs1 found that retinal function and structure of Rs1-KO mice were improved 3 months after injection of the vector. These findings show that the AAV-DJ (K137R) vector can efficiently and safely penetrate the inner limiting membrane and transduce different layers of retinal cells for a long time, while being able to continuously and efficiently express the target therapeutic protein, making it a candidate therapeutic vector for X-linked retinoschisis (XLRS).
The researchers first observed the time course of enhanced retinal green fluorescent protein (GFP) expression after intravitreal injection of AAV in mice. Images taken 14d\21d\28d\35d\3M\6M\12M after intravitreal injection clearly showed fluorescent expression in the retina, with the optic disc as the focal point. Among them, AAV-DJ-GFP and AAV-DJ (K137R)-GFP vectors showed GFP fluorescence expression on the 14th day after injection, and the fluorescence intensity reached a peak 35 days after injection. GFP fluorescence could still be detected 12 months after fluorescence. GFP expression in the retina injected with AAV8 was limited to the optic disc (Figure 1A). In contrast, after injection of AAV-DJ-GFP and AAV-DJ (K137R)-GFP, GFP signals accumulated in specific areas, such as around blood vessels covering Müller glial processes and around the optic disc (Figure 1B, C and D). Immunohistochemical analyses revealed GFP fluorescence expression in nerve fiber cells, horizontal cells, and Müller cells for AAV-DJ-GFP and AAV-DJ(K137R)-GFP (Figure 1F and G).
