| Summary | Creative Biogene ensures high-quality AAV particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between AAV particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in the production of AAV, especially for applications in animal studies and gene therapy. Effective endotoxin quality control is essential in the development and manufacturing of AAV particles. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced AAV particles to ensure regulatory compliance. |
| Purity | AAV purity is critical for ensuring the safety and efficacy of AAV-based applications.AAV capsids are composed of three main protein components, known as viral proteins: VP1, VP2, and VP3. These proteins play a critical role in the structure and functionality of the AAV capsid. Monitoring the VP1, VP2, and VP3 content in AAV preparations is essential for quality control in AAV production. Our AAV particles are tested for showing three clear bands of VP1, VP2 VP3 by SDS-PAGE. |
| Sterility | The AAV virus samples are inoculated into the cell culture medium for about 5 days to detect bacterial and fungal growth. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of AAV to deliver genetic material into target cells or tissues, and assess gene expression and functional activities. |
| Empty vs. Full Capsids | Based-on our proprietary AAV production and purification technology, Creative Biogene can always offer AAV particles with high ratio of full capsids. If required, we can also assess the ratio for a specifc lot of AAV particles by transmission electron microscopy (TEM) or other methods. |
Early studies used capsids and viral machinery derived from AAV serotype 2 (AAV2). AAV2 remains the basis for most AAV systems, but now, engineered capsids, including DJ and DJ8, have tissue-specific tropism or increased infectivity. DJ serotypes have also shown efficient transfection of many cultured cells, making DJ particularly suitable for cell culture and in vivo applications. In addition, specific promoters, the Cre/flox system, and gene editing via CRISPR have made AAV cell-specific and allowed for new approaches in gene therapy and animal studies.
Production of AAV requires cytopathogenicity, which occurs only after co-infection with a helper adenovirus or herpes virus. Helper viruses are difficult to remove and may cause adverse effects such as host inflammation. Current AAV expression systems avoid the use of helper viruses and instead use the plasmid pHelper, which contains essential genes such as E2A and E4. Human embryonic kidney (HEK) 293T cells expressing the SV40 large T antigen provide additional essential proteins. This AAV production system is known as the “helper-free” system and consists of three different plasmids encoding the essential viral and helper genes: pHelper, an AAV trans plasmid containing the AAV replication (Rep) and capsid (Cap) genes, and an AAV cis plasmid encoding the gene of interest, promoter, and inverted terminal repeats (ITRs).
The heart utilizes multiple adaptive mechanisms to maintain pump function. Compensatory cardiac hypertrophy reduces wall stress and oxygen consumption, thereby protecting the heart from acute increases in blood pressure. Yes-associated protein 1 (YAP), a nuclear effector of the Hippo pathway, is activated and mediates compensatory cardiac hypertrophy induced by acute pressure overload (PO). In this study, YAP promoted glycolysis by upregulating glucose transporter 1 (GLUT1), which in turn led to the accumulation of intermediates and metabolites of glycolytic, auxiliary, and supplementary pathways during acute PO. In the presence of acute PO, cardiac hypertrophy was suppressed and heart failure was exacerbated in YAP haploinsufficient mice. However, normalization of GLUT1 rescued the deleterious phenotype. PO induced the accumulation of glycolytic metabolites in a YAP-dependent manner, promoting cardiac hypertrophy. YAP upregulates the GLUT1 gene by interacting with TEA domain family member 1 (TEAD1) and HIF-1α in cardiomyocytes. Thus, YAP induces compensatory cardiac hypertrophy by activating the Warburg effect.
Here, researchers performed a GLUT1 rescue experiment, in which mice were injected with either AAV-DJ/8-GFP (AAV-control) or AAV-DJ/8-GLUT1 (AAV-GLUT1) and, after 2 weeks, subjected to either sham operation or TAC. Studies confirmed that AAV-GLUT1 significantly increased GLUT1 expression in the heart (Figure 1A and B). After 7 days of PO, mice were subjected to echocardiographic analysis and sacrificed. AAV-GLUT1 successfully rescued GLUT1 upregulation in YAPch-KO mice after 7 days of TAC (Figure 1C and D). Fractional shortening (FS, %), a measure of systolic function (Figure 1E and F), LV end-diastolic internal dimension (LVIDd), a measure of LV dilation (Figure 1E and G), and lung weight/tibia length (LungW/TL), a measure of lung congestion (Figure 1H), which is a sign of heart failure, were not affected in either AAV-control– or AAV-GLUT1–injected control mice. TAC-induced left ventricular systolic dysfunction and left ventricular dilation were observed in AAV control-injected YAPch-KO mice (Figure 1E-G). TAC-induced systolic dysfunction, left ventricular dilation, and pulmonary congestion were not evident in YAPch-KO mice injected with AAV-GLUT1 (Figure 1E-H).
