The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

Oral squamous cell carcinoma (OSCC) is a highly prevalent and aggressive malignancy characterized by a high risk of local invasion and recurrence. Interleukin-1α (IL-1α), a pro-inflammatory cytokine, has been implicated in the progression of various cancers, yet its specific role in OSCC remains unclear. In this study, researchers investigated the expression of IL-1α in human OSCC tissues and cell lines and analyzed its relationship with malignant cellular behaviors. The results revealed that IL-1α was highly expressed at the tumor invasive front and in clusters of budding cancer cells. Functional assays demonstrated that IL-1α knockdown significantly inhibited cancer cell migration but did not affect proliferation. These findings suggest that IL-1α promotes the invasive potential of OSCC and holds promise as a prognostic biomarker for the disease. Targeting the IL-1α signaling pathway may offer a novel therapeutic strategy to inhibit OSCC progression.
Here, the researchers investigated the effects of IL-1α expression on the HSC3 cell line. Twenty-four hours after siRNA transfection, both IL-1α mRNA expression and secretion levels in these cancer cells were reduced (Figure 1A, B). Subsequently, the study evaluated the impact of IL-1α knockdown on the cancer cell phenotype. The results showed that IL-1α knockdown did not significantly affect the proliferative capacity of the treated cells (Figure 1C). However, wound healing assays demonstrated that the migratory ability of the IL-1α-knockdown OSCC cell line was significantly inhibited (Figure 1D). These results suggest that suppressing IL-1α expression in cancer cells impairs their migratory capacity.
