Cervical cancer is one of the most common malignancies of the female reproductive system. Radioresistance is a significant factor limiting the efficacy of radiotherapy for cervical cancer. Previous studies have shown that interleukin-11 (IL-11) is highly expressed in various human cancers and is associated with clinical stage and poor prognosis. However, the specific role and underlying mechanisms of IL-11 in cervical cancer radioresistance remain unclear. In this study, analysis of the TCGA database revealed that IL-11 is highly expressed in cervical cancer tissues and correlates with clinical stage and poor patient prognosis. The researchers found that IL-11 levels were significantly elevated in radioresistant cervical cancer cells. Knockdown of IL-11 in HeLa cells reduced colony-forming survival and cell viability, induced G2/M phase arrest, and promoted apoptosis; conversely, exogenous addition of IL-11 to C33A cells increased colony-forming survival and cell viability while inhibiting G2/M phase arrest and apoptosis. Mechanistic studies indicated that IL-11-mediated radioresistance is associated with the activation of the PI3K/Akt signaling pathway. Collectively, these findings suggest that IL-11 contributes to radioresistance in cervical cancer and represents a potential target for radiosensitization therapy.
Increasing evidence indicates that IL-11 plays a pivotal role in tumorigenesis through three major signaling pathways: JAK-STAT3, RAS-Erk, and PI3K-Akt. Researchers used Western blot analysis to assess the levels of pSTAT3, pErk, and pAkt in HeLa cells with IL-11 knockdown. The results showed that Akt phosphorylation levels were significantly reduced in IL-11-knockdown HeLa cells; however, the phosphorylation levels of STAT3 and Erk remained unaffected (Figure 1A). Furthermore, the researchers examined changes in Bcl-2 (an anti-apoptotic protein), Bcl-xl (an anti-apoptotic protein), and Bax (a pro-apoptotic protein), which are downstream target genes of the Akt signaling pathway. Western blot results demonstrated that Bcl-2 and Bcl-xl protein levels were downregulated, whereas Bax protein levels were upregulated in IL-11-knockdown HeLa cells (Figure 1B). These findings suggest that reducing IL-11 levels may enhance the sensitivity of cervical cancer patients to radiotherapy by inhibiting the PI3K/Akt signaling pathway.
