Targeted LVV Process Development & GMP Manufacturing

Overview
Targeted lentiviral vectors (LVVs) introduce additional manufacturing considerations because engineered targeting components may influence vector stability, recovery, and functional performance during production and purification. Process conditions optimized for physical yield alone may not preserve targeting activity or transduction performance.
We provide process development and GMP manufacturing support for targeted LVV programs, covering upstream and downstream process development, scale-up, engineering runs, GMP production, and CMC support. Process development is guided by both manufacturing performance and the functional requirements of the vector.
Why Targeted LVV Manufacturing Is Different
Compared with conventional LVVs, targeted vectors may require additional process considerations related to their engineered surface components and functional properties.
1
Surface component stability
Engineered targeting moieties or modified envelope components may influence vector stability, recovery, and functional activity during downstream processing.
2
Yield–function tradeoff
Conditions that maximize physical vector recovery may not provide the best transduction efficiency or targeting specificity.
3
Narrow process windows
Changes in pH, shear conditions, or buffer composition may affect vector integrity and functional performance.
4
Analytical requirements
Conventional LVV assays such as p24 and qPCR provide important product information but do not fully assess targeting performance. Functional assays may therefore need to be incorporated into process development and characterization.
These considerations make it important to establish the manufacturing process alongside the specific vector architecture rather than relying solely on a standard LVV production workflow.
Integrated Development Strategy

Each phase is connected through defined process and analytical requirements to support a controlled transition from development to manufacturing.

GMP Manufacturing Capabilities
Following process development and scale-up, the established process can be transferred to GMP manufacturing to support clinical development of in vivo CAR-T programs.
Manufacturing is performed using qualified materials, controlled processes, and appropriate manufacturing systems, with process monitoring and documentation throughout production.
GMP Manufacturing Workflow:

- Comparability and bridging strategies to support process changes during clinical development
CMC & Regulatory Support
Integrated CMC support connects process development, manufacturing, and analytical characterization to support IND-enabling activities and clinical development of targeted LVVs.
Process Characterization
Evaluation of process performance and product attributes to support process understanding and control.
CQA / CPP Assessment
Identification and evaluation of critical quality attributes (CQAs) and critical process parameters (CPPs) to support process understanding, control strategy development, and manufacturing consistency.
Technology Transfer
Transfer of established process parameters, procedures, analytical methods, and manufacturing requirements into GMP manufacturing environments.
CMC Documentation
Preparation and compilation of process, manufacturing, and analytical information to support CMC documentation and regulatory activities.
From Targeted Vector Development to GMP Manufacturing
Moving a targeted LVV from early development to clinical supply requires the vector's functional properties to be considered alongside manufacturing performance. Our services support this transition by establishing processes that maintain product quality and functional performance as production moves toward clinical scale.
We work with in vivo CAR-T developers to establish targeted LVV manufacturing processes that are:
Scalable
Suitable for progression from preclinical batches to clinical-scale manufacturing
Reproducible
Supported by defined process parameters and batch-to-batch monitoring
Function-preserving
Designed to maintain targeting and transduction performance during processing and scale-up
Regulatory-aligned
Supported by appropriate analytical, process, and CMC documentation for clinical development
