In Vitro Transduction & Functional Assessment

OverviewAssessment FrameworkTechnologies & Cell SystemsWorkflowFeatures

Overview

Lentiviral vector-based in vivo CAR-T programs require a clear understanding of how candidate vectors perform in relevant immune-cell environments. Beyond transduction performance, our in vitro studies assess CAR expression across selected immune-cell populations, characterize the phenotype of CAR-expressing T cells, and evaluate antigen-dependent effector function.

We apply fit-for-purpose cellular and analytical methods to characterize lentiviral vector-mediated CAR expression and downstream immune-cell responses. Depending on study objectives, the assessment framework may integrate multiparameter flow cytometry, target-cell co-culture, cytotoxicity assays, cytokine and effector-response analysis, and molecular quantification.

Our preclinical CRO services generate integrated data on transduction, CAR expression, cellular phenotype, and functional activity. These data can support selection of study conditions and preparation for subsequent in vivo pharmacology, biodistribution, and safety evaluations.

In Vitro Transduction & Functional Assessment

Transduction and CAR Expression

This module assesses the ability of lentiviral vector candidates to mediate CAR expression in selected immune-cell systems.

Studies may be performed with peripheral blood mononuclear cells (PBMCs), enriched CD3+ T cells, CD4+ or CD8+ T-cell populations, or other project-relevant cell systems. Where constructs include reporter genes, reporter expression may be incorporated as a supporting transduction readout.

Assessment Endpoints

  • Frequency of CAR-positive cells
  • CAR expression intensity, including mean fluorescence intensity (MFI)
  • Reporter-positive cell frequency, where applicable
  • Post-transduction cell viability
  • Short-term cell recovery
  • CAR expression across selected post-transduction time points
  • Comparative transduction and expression performance across vector candidates or predefined vector input conditions

Applicable Methods

  • Multiparameter flow cytometry
  • CAR-specific detection assays
  • Tag-specific antibodies, Protein L, antigen probes, or project-specific detection reagents
  • Cell counting and viability assessment
  • Optional vector copy number (VCN) analysis by qPCR or ddPCR

We select the CAR detection strategy based on construct design, reagent availability, and sample characteristics. For CAR constructs without a broadly applicable detection reagent, assay suitability can be evaluated before the main study.

Immune-Cell Subset Distribution

This module characterizes the distribution of CAR expression or vector-associated signal across selected immune-cell subsets. It supports assessment of cell-population distribution patterns for lentiviral vector candidates in the selected in vitro immune-cell system.

The flow cytometry panel is configured according to the study objective, sample type, and relevant cell populations.

Cell Populations

  • CD3+ T cells
  • CD4+ T cells
  • CD8+ T cells
  • B cells
  • NK cells
  • Monocytes and other project-relevant immune-cell populations
  • Regulatory T-cell-associated populations, where required

Key Readouts

  • CAR-positive cell frequency within each selected immune-cell subset
  • Relative distribution of CAR-positive cells across immune-cell subsets
  • CAR expression across CD4+ and CD8+ T-cell populations
  • Relative CAR distribution between T-cell and selected non-T-cell populations
  • Comparative subset-distribution profiles across vector candidates

We interpret these results in the context of the tested vector, cell source, culture conditions, selected time points, and control design. The resulting dataset provides an in vitro view of cell-subset-associated CAR expression and complements downstream in vivo studies.

CAR-Expressing T-Cell Phenotype

This module evaluates the phenotype of CAR-expressing T cells to characterize cellular status following lentiviral vector transduction. The analysis can provide data on viability, activation, differentiation-associated features, and selected dysfunction-associated markers.

Flow cytometry panels may be configured as focused characterization panels or expanded according to project objectives.

Core Phenotypic Endpoints

  • CD4/CD8 composition
  • Cell viability
  • T-cell activation-associated markers
  • Comparison of phenotypic features between CAR-positive and CAR-negative cell populations

Optional Extended Endpoints

  • Activation markers: CD25, CD69, HLA-DR
  • Differentiation-associated markers: CD45RA, CCR7, CD62L, CD27, CD28
  • Dysfunction/exhaustion-associated markers: PD-1, TIM-3, LAG-3, TIGIT
  • Cell proliferation-related parameters
  • Additional markers relevant to the CAR mechanism or project objectives

Phenotypic data can be integrated with CAR expression, target-cell killing, and cytokine-release results to provide a consolidated characterization of the tested CAR-expressing cell population.

Antigen-Specific Cytotoxicity

This module evaluates the ability of CAR-expressing effector cells generated after lentiviral vector transduction to kill target cells under defined co-culture conditions.

We design cytotoxicity studies with antigen-positive target cells, antigen-negative or low-expression control cells, and relevant effector-cell comparators. This approach supports assessment of target-cell killing in relation to target-antigen expression and background activity.

Typical Study Elements

  • Target-antigen-positive target cells
  • Antigen-negative or low-expression control cells
  • Defined effector-to-target (E) ratios
  • Single or multiple co-culture time points
  • CAR-expressing, untransduced, and mock-treated effector-cell controls
  • Comparative assessment of multiple lentiviral vector candidates
  • Feasibility assessment for customer-provided target cells

Key Readouts

  • Target-cell viability
  • Specific lysis
  • Target-cell clearance
  • Cytotoxic response across selected E:T ratios
  • Killing response at selected time points
  • Differences between antigen-positive and antigen-negative target-cell conditions

Depending on target-cell characteristics, assay duration, and intended endpoints, cytotoxicity may be assessed using flow cytometry, fluorescenceor luminescence-based methods, cell counting, or real-time cell analysis.

Cytokine Release and Effector Response

This module assesses cytokine release and effector-function-associated responses following co-culture of CAR-expressing cells with target cells. These assays complement cytotoxicity assessments by characterizing cellular responses under antigen-stimulation conditions.

We configure the cytokine panel based on the target antigen, CAR signaling architecture, target-cell model, co-culture duration, and project requirements.

Commonly Assessed Analytes

  • IFN-γ
  • IL-2
  • TNF-α
  • GM-CSF
  • Granzyme B
  • Perforin
  • IL-6 and other project-relevant inflammation-associated analytes
  • CD107a degranulation signal
  • Intracellular cytokine expression, where applicable

Key Readouts

  • Cytokine concentrations following co-culture with antigen-positive target cells
  • Differences relative to antigen-negative target-cell or untransduced-cell controls
  • Comparative effector-response data across vector candidates
  • Association of cytokine-release results with CAR expression or cytotoxicity data
  • Degranulation and intracellular effector-molecule readouts

Cytokine and effector-function analysis can be performed using ELISA, multiplex cytokine platforms, flow cytometry, and other fit-for-purpose analytical approaches.
