Targeted Lentiviral Vector Engineering

OverviewStrategyApplicationFeatures

Overview

Targeted lentiviral vector (LVV) engineering enables selective delivery of genetic payloads to defined immune-cell populations, providing an important strategy for in vivo CAR-T cell development. Unlike broadly tropic VSV-G-pseudotyped LVVs, targeted LVVs are engineered to preferentially recognize selected cell-surface receptors while reducing non-target cell transduction.

We provide integrated targeted LVV engineering support spanning target selection, targeting ligand design, envelope and fusogen engineering, vector architecture optimization, and targeting performance evaluation. Targeting strategies can be tailored to specific immune-cell populations, including T-cell subsets, using receptor-directed binders and engineered viral envelopes. Candidate vectors can then be evaluated for targeting specificity, transduction efficiency, selectivity, and functional CAR expression to support the identification of LVV candidates for downstream in vivo evaluation.

Targeted LVV Engineering Strategy

Targeted LVV development requires coordinated optimization across the entire delivery system:

Target Recognition

Selective Entry

CAR Delivery

Functional Expression

Targeting Validation

We integrate these engineering steps rather than optimizing individual components in isolation. Target receptor characteristics guide ligand selection, ligand configuration informs envelope engineering, and the resulting targeting system is evaluated together with the CAR vector architecture.

This integrated approach enables systematic identification of limiting factors and supports iterative refinement of targeted LVV candidates.

01 — Target Selection & Ligand Engineering

Target receptor selection establishes the basis for cell-selective LVV delivery. We assess receptor expression, accessibility, and distribution across target and non-target cell populations to determine whether a receptor is suitable for targeted vector development.

We then select or engineer an appropriate receptor-binding ligand and optimize its configuration for incorporation into the LVV targeting system.

Engineering Focus

Target Receptor Assessment

Expression • Accessibility • Target/Non-Target Distribution • Receptor Suitability

Potential Targeting Modalities

scFv • VHH/Nanobody • DARPin • Fab • Other receptor-binding ligands

Targeting Configuration

Binding Affinity • Ligand Orientation • Display Configuration • Receptor Accessibility

02 — Envelope & Fusogen Engineering

The envelope is a central determinant of LVV tropism and cell entry. For targeted delivery, we engineer the envelope system to reduce undesired native receptor interactions while introducing mechanisms for preferential recognition and entry into the intended cell population.

Our approach can incorporate native receptor detargeting, targeting ligand display, envelope/fusogen selection, and fusion optimization according to the target receptor and vector design.

Engineering Focus

  • Native receptor detargeting
  • Targeting ligand display
  • Envelope and fusogen selection
  • Fusion activity optimization
  • Envelope stability and vector performance
  • Compatibility with downstream vector production

Targeted Entry Strategy

01 Reduce Native Entry:

Minimize undesired envelope-mediated transduction of non-target cells.

02 Introduce Receptor-Specific Recognition:

Incorporate and optimize a targeting ligand for the intended cell-surface receptor.

03 Maintain Productive Fusion Activity:

Preserve efficient membrane fusion, vector integrity, and functional titer.

04 Evaluate Target-Cell Selectivity:

Compare transduction and CAR expression in target-positive and non-target cell models.

Envelope engineering is performed together with ligand and vector architecture optimization to establish a targeted LVV configuration suitable for subsequent functional evaluation.

03 — Vector Architecture & Payload Optimization

Selective entry alone is insufficient for effective in vivo CAR-T generation. The vector genome must also support appropriate CAR expression while maintaining overall vector performance.

We optimize the CAR expression cassette and regulatory architecture in parallel with the targeting system, considering promoter selection, regulatory elements, payload configuration, and strategies for controlling unwanted expression.

Key Optimization Areas

  • CAR cassette configuration
  • Promoter and regulatory element selection
  • Payload size and vector performance
  • Regulatory sequence optimization
  • miRNA-based detargeting strategies
  • Production-Stage Payload Control

04—Targeting Specificity & Transduction Evaluation

Targeted LVV candidates are evaluated using comparative models to determine whether delivery is preferentially associated with the intended receptor and target-cell population.

Target-positive and target-negative cells are assessed in parallel, with receptor-blocking, receptor-deficient, or other mechanistic controls incorporated where appropriate.

Targeting Efficiency

  • Target-cell transduction
  • CAR expression

Targeting Specificity

  • Target vs non-target transduction
  • Selectivity ratio

Receptor Dependence

  • Receptor blocking
  • Receptor-deficient models

Vector Performance

  • Functional titer
  • Particle characteristics
  • Vector integrity

Functional Activity

  • CAR expression
  • T-cell activation
  • Cytotoxicity

The resulting data are used to determine whether targeting performance is primarily limited by ligand recognition, envelope-mediated entry, vector architecture, or downstream CAR expression, enabling focused optimization rather than trial-and-error engineering.
