Cell Line DescriptionThe JeKo-1 mantle cell lymphoma (MCL) cell line was isolated from peripheral blood mononuclear cells of a 78-year-old woman with large-cell type mantle cell lymphoma that had undergone leukemic transformation. JeKo-1 cells were Epstein-Barr virus negative and showed a B-cell phenotype with IgM+, IgD+, CD3−, CD5+, CD10−, CD19+, CD20+ and CD23−. They overexpressed cyclin Dl, Bcl-2, c-Myc and Rb proteins. JeKo-1 cells showed complex chromosomal aberrations, but cytogenetically lacked an apparent t(11;14)(q13;q32) chromosomal translocation. However, bcl-1/JH gene rearrangement was confirmed at the molecular level, and overexpression of PRAD1/cyclin D1 protein was revealed in JeKo-1 cells and the original BM lymphoma cells. The JeKo-1 cells are highly tumorigenic in SCID mice and have applications for immunology research.

Additional Infop53 mutations and protein overexpression have been associated with poor prognosis or blastic variants of MCL. A substantial number of JeKo-1 cells were large or anaplastic in morphology, and the original tumour showed an aggressive clinical course, but JeKo-1 cells did not overexpress p53 protein. JeKo-1 cells expressed Rb, c-Myc and Bcl-2 instead. Zuckerberg et al demonstrated that most cases of MCL express both cyclin Dl and Rb. Bcl-2 has been shown to be consistently expressed in MCL. These results were consistent with the present data.

Culture Conditions and Handling
SubculturingHomogenize the cell suspension in the flask by gently pipetting up and down, then take a representative sample to determine the cell density per milliliter. Dilute the suspension with fresh medium to a concentration of 1 x 10^5 cells/mL and aliquot the adjusted suspension into new flasks for continued culture.
