microRNA mimics are double-stranded miRNA-like RNA that are designed to copy the functionality of mature endogenous miRNA upon transfection. Once these RNA fragments are introduced into cells, the miRNA mimics can bind specifically to the targeted gene. The result is posttranscriptional repression or more specifically translational inhibition of the gene. miR-mimics will act in a gene-specific fashion allowing for “miRNA-targeting” and “miRNA-gain-of-function” methods that are presently primarily used as exogenous tools to study gene function by targeting mRNA through miRNA-like actions in mammalian cells.
microRNA inhibitors are complementary, antisense single-stranded oligonucleotides to their target, endogenous mature miRNA. Synthetic miRNA inhibitor designs incorporate the reverse complement of the mature miRNA (the target site) and are chemically modified to prevent RISC-induced cleavage, enhance binding affinity and provide resistance to nucleolytic degradation. When delivered to a cell, binding of endogenous mature miRNAs to these complementary synthetic target sites is thought to be irreversible, therefore these inhibitors are presumed to sequester the endogenous miRNA, making it unavailable for normal function.
