Creative Biogene offers filter-binding radiometric kinase activity assay, which has long been established as the most reliable, robust and gold standard assay against all other assay formats. We can also provide TR-FRET technology to measure substrate phosphorylation when a greater level of throughput is desired to support high throughput screens. Radiometric based assays are the most reliable and stable methods for detecting kinase reactions; therefore, they are the preferred method for kinase profiling assay. Compared with those nonradiometric methods, it is considered the radiometric based filtration binding assay as the ‘gold standard’ method. In this assay format, the kinase reaction is carried out in the presence of 32P-γ-ATP or 33P-γ-ATP. By binding of the final radioisotope labeled products to filters, unreacted phosphate can be washed away without interfering with the detection of real phosphorylated products (Haiching, et al. 2008). In addition, scintillation proximity assay (SPA) is also used in radiometric kinase activity test.

The FRET assay is a kind of technology that is amenable to automation. This assay uses ‘donor’ and ‘acceptor’ fluorophores in proximity, with the excited donor fluorophore exhibiting nonradiative energy transfer to an acceptor fluorophore (Haiching, et al. 2008).
